If you just start on quest 21-1 and just play through the rest of the quests on super your most likely going to get a good amount of gems. Or you can just do 21-1 on super over and over and try to get Pilaf and Launch to appear over and over. Pilaf gives 10 and Launch 15.

If you're not worried about ranking up (like when farming when 3x Exp. isn't on) then I'd honestly suggest sticking with 21-1 or 18-3.If you only care about gems, the very last stage is pretty good for quickly seeing if you're gonna get gems or not.
